100 years old will be the new 60

For the first time in history, leading scientists and entrepreneurs believe there’s a way to slow aging — and maybe even reverse it.The latest research on longevity suggests there is no reason that people born today can’t live to at least 120 years old… perhaps even to 150 and beyond.How would you change your life if you could live to 120 years old and remain healthy?What would you do differently today?Last week we explored the “Longevity Mindset.” Today and next week, I’ll do a quick review of the latest advancements toward rewiring the biology of aging. Every year, I take a group of my Abundance 360 Members on a “Platinum Longevity Trip” to meet with the cutting-edge researchers and companies. Following are some of the companies and technologies we observed that have the potential to increase your healthspan — the amount of time you have to live a healthy and functional life, avoiding expensive end-of-life care.

Over the past two decades, the cost of sequencing the human genome has dropped 100,000-fold: from $100 million per genome, to below $1,000 per genome (current estimates are as low as $300)
Genome sequencing can uncover disease susceptibilities years before symptoms present, allowing for personalized preventative care to begin sooner than ever before. 
For example, the Cancer Genome Atlas Program at the NIH is currently using gene sequencing to decode the genetic underpinnings of 30 cancer types.
Perhaps the most impactful potential of low-cost genome sequencing is its ability to be used in what is called a liquid biopsy — the ability to find free-flowing cancer DNA in your bloodstream that might indicate the existence of an undetected cancer in your body.  And, as we know, finding cancer at stage-zero or stage-one is the key to survival. 
There are two major companies we visited with during our Platinum Longevity Trip:
Cancer detection company GRAIL analyzes the mutated, fractionated DNA and RNA from cancer cells in your blood (from a simple blood draw) to diagnose over 50 cancer types in early stages. GRAIL recently received an $8 billion buy-out offer from biotech giant Illumina.
Freenome takes a similar approach to early cancer diagnosis from a real-time blood draw (called a liquid biopsy), initially focused on colon cancer. Freenome’s multiomics platform analyzes fragments of DNA, RNA, and protein from the cancer and from the host response. This form of precision medicine bridges early detection and early intervention to boost human healthspan. 

Gene Therapy
One of the most powerful technologies now available in the fight for longevity is called gene therapy — a technology theorized in the 1980s that has taken almost 40 years to mature. Gene therapy allows scientists to use a vector (typically an Adeno Associated Virus) to carry a desired gene to a set of desired cells in an organism. Want a specific gene put into retinal cells, or bone marrow, or neurons? No problem, there’s a gene therapy approach for that.
A new biotech start-up called Gordian Biotechnology is using the convergence of gene therapy and single cell sequencing to run hundreds of thousands of independent experiments in a single animal to determine the therapeutic effects of specific gene additions on specific cells of interest. Because aging is such a multifactorial process, this approach can run thousands of parallel experiments to tackle the many complexities of age-related diseases simultaneously. 
Next week, we’ll learn about a company called Rejuvenate Bio, and an extraordinary researcher named Dr. David Sinclair who is using gene therapy to potentially rejuvenate animals with the ultimate goal of age reversal in humans.

CRISPR: Cut, Paste, and Search Your Genes 
In addition to Gene Therapy, the other incredible tool in our longevity research arsenal is CRISPR. 
You may know CRISPR as the molecular scissors that can edit genes — think CTRL X (cut) and CTRL V (paste). But beyond cutting and pasting, CRISPR can also be used to help find and identify a sequence of DNA in your cell, sort of a “CTRL F” functionality. This discovery is so important and transformative that the Nobel Prize was just awarded this month to Dr. Jennifer Doudna of Gladstone Institutes for its discovery.
Here are several other exciting CRISPR developments:

Gladstone-UCSF Institute of Genomic Immunology is using CRISPR to edit the T-cells of the immune system that play critical roles in cancer, infection, and autoimmunity. CRISPR can delete mutated genes or add new DNA to reprogram the T-cells. This personalized approach takes advantage of the body’s own immune system to tackle complex diseases. 
Rather than snipping and replacing genes, Mammoth Biosciences has programmed CRISPR proteins to locate and cleave target genes, acting as “molecular shredders.” The cleaved gene serves as a molecular readout if the target is successfully bound, enabling CRISPR to serve as a diagnostic tool. Additionally, the company’s novel CRISPR proteins (cas14, casV) exist in the micro and nano scales, opening the door for new delivery systems at smaller scales than ever before.
With the recent breakthroughs in CRISPR and Gene Therapy technologies, a variety of strategies for reversing disease have been tried. Yet countless experiments remain, and that’s where AI can help.

Artificial Intelligence: Catalyzing Discovery 
The explosion of novel imaging, sensing, and sequencing tools has unleashed an abundance of patient data. 
But bringing together this information across millions of patients to form actionable insights can only be achieved with Artificial Intelligence. 
One of the leading companies in this area is Insilico Medicine, which is leveraging AI in its end-to-end drug pipeline, extending healthy longevity through drug discovery and aging research. 
In their comprehensive drug discovery engine, Insilico uses millions of samples and multiple data types to A) discover signatures of diseases, and B) identify the most promising targets for billions of molecules. These molecules either already exist or can be generated de novo with the desired set of parameters.
Insilico uses an AI technique called generative adversarial networks (GANs) to “imagine” novel molecular structures. With reinforcement learning, Insilico’s system lets you generate a molecule with any of up to 20 different properties to hit a specified target.
Thanks to converging breakthroughs in machine learning, drug discovery and molecular biology, companies like Insilico can now do with 50 people what the pharmaceutical industry can barely do with an army of 5,000.
Another extraordinary company on the Longevity Platinum Trip was a company out of the Buck Institute called Edifice Health, which has developed the ability to determine your “inflammatory age” using advanced AI to score biomarkers of immune health. Inflammation is a leading contributor to most chronic illnesses, and greater awareness of this symptom will enhance preventative care. Even more important than measuring inflammatory age, Edifice Health is screening thousands of molecules to determine which can quell such inflammation.
An additional company out of the Buck Institute is Gerostate Alpha, a pharmaceutical company that is using large-scale AI to test millions of compounds for their ability to extend the life of a worm-like creature called the nematode. Once they get a hit in nematodes (rather short-lived creatures), they will then test the molecules in mice and eventually in humans. The company is testing millions of compounds in parallel, hoping to literally discover the pharmaceutical fountain of youth.

The 9 Mechanisms of Aging:?

*This section was adapted from CB INSIGHTS: The Future Of Aging.Longevity, healthcare, and aging are intimately linked.With better healthcare, we can better treat some of the leading causes of death, impacting how long we live.By investigating how to treat diseases, we’ll inevitably better understand what causes these diseases in the first place, which directly correlates to why we age.Following are the nine hallmarks of aging. I’ll share examples of health-tech and longevity technologies addressing each of these in coming weeks.Genomic instability: As we age, the environment and normal cellular processes cause damage to our genes. Activities like flying at high altitude, for example, expose us to increased radiation or free radicals. This damage compounds over the course of life and is known to accelerate aging.Telomere attrition: Each strand of DNA in the body (known as chromosomes) is capped by telomeres. These short snippets of DNA repeated thousands of times are designed to protect the bulk of the chromosome. Telomeres shorten as our DNA replicates; if a telomere reaches a certain critical shortness, a cell will stop dividing, resulting in increased incidence of disease.Epigenetic alterations: Over time, environmental factors will change how genes are expressed, i.e., how certain sequences of DNA are read and the instruction set implemented.Loss of proteostasis: Over time, different proteins in our body will no longer fold and function as they are supposed to, resulting in diseases ranging from cancer to neurological disorders.Deregulated nutrient-sensing: Nutrient levels in the body can influence various metabolic pathways. Among the affected parts of these pathways are proteins like IGF-1, mTOR, sirtuins, and AMPK. Changing levels of these proteins’ pathways have implications on longevity.Mitochondrial dysfunction: Mitochondria (our cellular power plants) begin to decline in performance as we age. Decreased performance results in excess fatigue and other symptoms of chronic illnesses associated with aging.Cellular senescence: As cells age, they stop dividing and cannot be removed from the body. They build up and typically cause increased inflammation.Stem cell exhaustion: As we age, our supply of stem cells begins to diminish as much as 100 to 10,000-fold in different tissues and organs. In addition, stem cells undergo genetic mutations, which reduce their quality and effectiveness at renovating and repairing the body.Altered intercellular communication: The communication mechanisms that cells use are disrupted as cells age, resulting in decreased ability to transmit information between cells.ConclusionOver the past 200 years, we have seen an abundance of healthcare technologies enable a massive lifespan boom.Now, exponential technologies like artificial intelligence, 3D printing and sensors, as well as tremendous advancements in genomics, stem cell research, chemistry and many other fields, are beginning to tackle the fundamental issues of why we age. In next week’s blog, we will dive into how genome sequencing and editing, along with new classes of drugs, are augmenting our biology to further extend our healthy lives.What will you be able to achieve with an extra 30 to 50 healthy years (or more) in your lifespan? AI, Quantum & Entrepreneurship in China

Last week, Eric Schmidt, Chairman of Alphabet, predicted that China will “rapidly overtake the U.S. in artificial intelligence… in as little as five years.”

Last month, China announced plans to open a $10 billion quantum computing research center in 2020.

Bottom line, China is aggressively investing in exponential technologies, pursuing a bold goal of becoming the global AI superpower by 2030.

Based on what I’ve observed from China’s entrepreneurial scene, I believe they have a real shot of hitting that goal.

As I described in a previous tech blog, I recently traveled to China with a group of my Abundance 360 members, where I was hosted by my friend Kai Fu Lee, the founder of Sinnovation Ventures.

On one of our first nights, Kai Fu invited us to a special dinner at DaDong Roast, which specializes in Pei King duck, where we shared an 18-course meal.

The meal was amazing, and Kai Fu’s dinner conversation provided us priceless insights on Chinese entrepreneurs.

Three topics opened my eyes. Here’s the wisdom I’d like to share with you.

  1. The Entrepreneurial Culture in China

Chinese entrepreneurship has exploded onto the scene and changed significantly over the past 10 years.

IMHO, one significant way that Chinese entrepreneurs vary from their American counterparts is in work ethic. The mantra I found in the startups I visited in Beijing and Shanghai was “9-9-6” – meaning the employees only needed to work from 9 am to 9 pm, 6 days a week.

Right now, online platforms like Khan Academy and Coursera have made a plethora of educational resources available 1) for free and 2) on demand, such that you can pick and choose what you learn and at what speed you learn it.

Khan Academy has delivered over 300 million lessons since it started in 2006, features 5,000 free instructional videos in 65 languages, and allows users to complete 4 million exercises every day – simply amazing.

Can a company actually disrupt itself?

I often say that businesses must disrupt themselves (before someone else does) to survive.

The fact of the matter is: very few companies have actually successfully disrupted themselves.

Instead, most successful companies “disrupt adjacencies”: they leverage their existing assets to expand into new, high growth markets. They actually disrupt someone else!

Let’s start with a few disruption examples.

FACEBOOK Disrupts SMS MESSAGING: Facebook decided to disrupt SMS messaging with the launch of Messenger. Because Facebook is a platform, they have been able to garner 700M monthly active users globally – driving a projected 38% decline in Telco SMS revenue in North America by 2017.

TESLA Disrupts ENERGY STORAGE: Tesla, an electric car company, is disrupting energy storage with the Tesla Powerwall. They used the technology developed for their cars to branch into this new ($19 billion) market.

GOOGLE disrupts MOBILE PHONES: Google is an Internet search company, but in 2008, Google got into the phone/hardware business, shipping Android and beginning the disruption of mobile operating systems. Android currently commands an astounding 82.8% market share.

Humanity is moving from evolution by natural selection (Darwinism) to evolution by intelligent direction.

For most of human history, our average age was only about 26 years old.

We would procreate at age 13, live just long enough to help our children raise their children, and then, on average, die at age 26 (so we were no longer taking food from the mouths of our grandchildren).

It was through technological innovation — sanitation and germ theory — that we moved life expectancy from 26 to the mid 50’s. Recently, because of modern medicine’s progress in treating heart disease and cancer, we’ve bumped up today’s global average human lifespan to 71 years.

But this is just the beginning.

Advances over the next 10 to 15 years will move life expectancy north of 100.

Reading – Sequencing the Human Genome

Your genome is the software that runs your body.

It is composed of 3.2 billion “letters,” or base pairs, that code foreverything that makes you “you” – your hair color, your height, your personality, your propensity to disease, your lifespan, and so on.

Until recently, it’s been very difficult to rapidly and cheaply “read” these letters and even more difficult to understand what they do.

In 2001, my friend and Human Longevity Inc. co-founder Dr. J. Craig Venter sequenced the first complete human genome. It took about a year and cost $100 million.

Since then, the cost to sequence a genome has been plummeting exponentially, outpacing Moore’s Law by almost 3x (take a look at the graph below).

Today, the cost to sequence a full human genome is about $1,000.

The World in 2025: 8 Predictions for the Next 10 Years

In 2025, in accordance with Moore’s Law, we will see an acceleration in the rate of change as we move closer to a world of true abundance.

Here are eight areas where we’ll see extraordinary transformation in the next decade:

A $1,000 Human Brain: In 2025, $1000 should buy you a computer able to calculate at 10^16 cycles per second (10,000 trillion cycles per second), the equivalent processing speed of the human brain.

A Trillion-Sensor Economy: The Internet of Everything describes the networked connections between devices, people, processes and data. By 2025, the IoE will exceed 100 billion connected devices, each with a dozen or more sensors collecting data. This will lead to a trillion-sensor economy driving a data revolution beyond our imagination. Cisco’s recent report estimates the IoE will generate $19 trillion of newly created value.

Rising Billions – dramatic (positive) change?

The most dramatic (positive) change in our global economy is about to occur between 2016 and 2020.

Three to five billion new consumers, who have never purchased anything, never uploaded anything, and never invented and sold anything, are about to come online and provide a mega-surge to the global economy.

While most of these individuals are in Africa, India, China and the developing world, and their income is low, when aggregated, this represents tens of trillions of new dollars flowing into the global economy… and no one is talking about it.

I call this group the “Rising Billions.”
